x-mod on "Browser Codes 404 & 500"

I'm using multisite and can't understand what's going on. I have followed the instructions to a "T" a the codex.wordpress.org site.
The problem is when I add the rewrite code into the .htaccess I get the 500 error but when I remove it I get 404 not found. Any help would be great.
